One area where it has made major inroads is medicine, biotechnology and pharmacology, where it is transforming <a href=\"https:\/\/venturebeat.com\/ai\/how-protais-ai-powered-platform-is-improving-drug-discovery\/\">drug discovery<\/a> and development.<\/p>\n<p>The <a href=\"https:\/\/www.lshtm.ac.uk\/newsevents\/news\/2020\/average-cost-developing-new-drug-could-be-15-billion-less-pharmaceutical\" target=\"_blank\" rel=\"noreferrer noopener\">cost<\/a> of discovering and developing a drug averages $1.3 billion and \u201cdemands anything from 12 to 15 years to hit the market,\u201d according to a <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/21091654\/\" target=\"_blank\" rel=\"noreferrer noopener\">PubMed<\/a><a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/21091654\/\"> paper<\/a>. So it\u2019s not surprising that the drug discovery industry has seen a significant rise in AI-powered technologies. A case in point is a paper in <a href=\"https:\/\/www.nature.com\/articles\/d41573-022-00025-1\" target=\"_blank\" rel=\"noreferrer noopener\">Nature<\/a> that notes that the integration of AI into the drug discovery and development pipeline has increased almost 40% annually.<\/p>\n<p>According to healthcare investors Tzvi Bessler and Morris Laster, Ph.D., \u201cdrug discovery companies are leveraging AI in a variety of ways, such as using machine learning algorithms to identify potential drug candidates, predict their effectiveness and safety, and optimize their design. Here are three trends that stood out:<\/p>\n<h2 id=\"h-1-more-efficiency-in-biology-modeling-and-drug-target-discovery\"><strong>1. More efficiency in biology modeling and drug target discovery<\/strong><\/h2>\n<p>James Handler, professor at Rensselaer Polytechnic Institute and chair of the Association for Computing Machinery Technology Policy Council, told VentureBeat about two uses where AI is showing great promise in drug discovery: reducing the number of potential candidates for trials, and providing potential explanations for the secondary use of drugs \u2014 that is, why a drug shows efficacy for a condition it wasn\u2019t originally designed to treat.<\/p>\n<p>In both cases, he noted, the key is that \u201cAI can reduce the number of possibilities that need to be explored through traditional means.\u201d This aids in biology modeling and drug target discovery. \u201cHowever,\u201d he added, \u201can important aspect of this is the AI systems being able to explain their predictions to humans, a focus of current <a href=\"https:\/\/arxiv.org\/abs\/2204.06614\" target=\"_blank\" rel=\"noreferrer noopener\">research<\/a>. This allows humans to make the final decisions [on] analysis and testing, with AI drastically reducing the cost of getting successful drugs to market.\u201d<\/p>\n<p>Drug discovery and development typically begins with identifying a biological target \u2014 a gene, protein, receptor or enzyme, for example. Proteins are the <a href=\"https:\/\/www.longdom.org\/open-access\/proteins-as-biomolecular-drug-targets-60942.html\">most <\/a><a href=\"https:\/\/www.longdom.org\/open-access\/proteins-as-biomolecular-drug-targets-60942.html\" target=\"_blank\" rel=\"noreferrer noopener\">common<\/a><a href=\"https:\/\/www.longdom.org\/open-access\/proteins-as-biomolecular-drug-targets-60942.html\"> drug targets<\/a> because of their ability to influence a cell\u2019s behavior or function. So traditional drug discovery efforts have involved selecting specific proteins with pockets that can be influenced by promising drug-like molecules (which then become the ligand, or binding drug).<\/p>\n<p>However, this process is computationally challenging. Of the 20,360 human proteins stored in the <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/9016499\/#:~:text=SWISS%2DPROT%20is%20a%20curated,of%20integration%20with%20other%20databases.\" target=\"_blank\" rel=\"noreferrer noopener\">SWISS<\/a><a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/9016499\/#:~:text=SWISS%2DPROT%20is%20a%20curated,of%20integration%20with%20other%20databases.\">-PROT<\/a> \u2014 the world\u2019s most widely-used, expertly curated protein sequence database \u2014 only a few have been explored as drug targets.<\/p>\n<p>Organizations are now using AI\u2019s ability to correlate and match large amounts of data, leading to more efficient drug target identification and discovery. In 2022, many AI-powered healthcare enterprises channeled resources toward building advanced modeling tools that not only model biology but also identify and validate new targets. This year, major pharmaceutical enterprises like AstraZeneca and Pfizer partnered with AI vendors that offer target discovery-as-a-service to <a href=\"https:\/\/www.biopharmatrend.com\/post\/615-pharmaceutical-artificial-intelligence-key-developments-in-2022\/#section-4\">discover over eight new targets<\/a>.<\/p>\n<h2 id=\"h-2-improved-protein-structure-prediction\"><strong>2. Improved protein structure prediction<\/strong><\/h2>\n<p>Proteins need to be folded into specific three-dimensional structures. Incorrect or absent folding has been <a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/B9780123971548001810\">linked<\/a> to the pathology of many diseases. Predicting protein structure is also relevant in the drug discovery process because it provides a better understanding of how the protein works, thereby informing how it can be affected, controlled and modified.<\/p>\n<p>This is a difficult task, however. One computational biology <a href=\"https:\/\/www.pnas.org\/doi\/10.1073\/pnas.1914677117\">research report<\/a> noted that protein structure prediction \u201cremains a prevailing challenge.\u201d<\/p>\n<p>However, 2022 inspired significant progress in predicting how proteins fold. This was spearheaded by DeepMind\u2019s innovative <a href=\"https:\/\/venturebeat.com\/2021\/06\/18\/what-is-open-source\/\">open-source<\/a> software, <a href=\"https:\/\/venturebeat.com\/business\/deepmind-open-sources-alphafold-2-for-protein-structure-predictions\/\">AlphaFold<\/a>, which can predict a protein\u2019s 3D structure from its one-dimensional amino acid sequence. AlphaFold was able to predict the protein structures of \u201cnearly all cataloged proteins known to science.\u201d<\/p>\n<p>Reducing what would typically take years to mere seconds, in July the software used the power of AI\u2019s deep learning to predict and publicly share over 200 million protein structures belonging to animals, plants, bacteria, fungi and other organisms.<\/p>\n<p>In November, DeepMind\u2019s AI model found a worthy rival in Meta\u2019s research team. Meta leveraged AI\u2019s natural language processing (NLP) abilities and applied \u201ca large language model\u201d to predict the structure of over 600 million proteins found in both known and unknown organisms. As de novo is typically a <a href=\"https:\/\/venturebeat.com\/ai\/how-2022-became-the-year-of-generative-ai\/\">generative<\/a> type of design, it relies largely on computational processes and <a href=\"https:\/\/venturebeat.com\/ai\/10-years-on-ai-pioneers-hinton-lecun-li-say-deep-learning-revolution-will-continue\/\">deep learning<\/a> models.<\/p>\n<p>2022 has witnessed significant progress in the development of de novo approaches that incorporate <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/34731479\/\">reinforcement-<\/a><a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/34731479\/\" target=\"_blank\" rel=\"noreferrer noopener\">learning<\/a><a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/34731479\/\"> architectures<\/a> in regular AI <a href=\"https:\/\/venturebeat.com\/2022\/03\/27\/this-is-what-makes-deep-learning-so-powerful\/\">neural networks<\/a>.<\/p>\n<p>The virtual screening of existing databases, another aspect of drug design, was also an object of attention in 2022. Combing through large databases for similarities and spotting specific peculiarities are defining features of AI. Pharmaceutical giants applied this technology to large volumes of databases and invested millions of dollars in partnerships with AI platforms capable of virtually screening trillions of synthesized compounds.<\/p>\n<p>Handler noted that drugs that seem to be effective in animal testing often fail when they reach human trials. The challenge is predicting toxicity from the earlier data, he said. \u201cNew techniques are exploring how to use AI models that integrate the many kinds of test data to predict toxicity better and consequently reduce the number of candidates needing expensive testing.\u201d<\/p>\n<p>Handler added that more data is becoming available and shared, and <a href=\"https:\/\/venturebeat.com\/ai\/6-healthcare-ai-predictions-for-2023\/\">predicts<\/a> that \u201cthis should create many opportunities for innovation in drug discovery\u201d going into 2023.
